Tissue Culture, Have You Had Success?
Josh Young (Thu, 09 Sep 2010 17:19:49 PDT)

This is a very interesting topic, how many of you have tried this and been
successful?  Honestly, I wouldn't have ever assumed it to be succesful outside
of a sterile lab.

Just a further note on sterility.  It doesn't hurt to keep a clean set of
clothes in the room dvoted to the tissue culture.  And to spray it with a dilute
solution of Clorox or Physan prior to use. Keep tall est tubes or vials with
cotton or other plugs in them in the room and sterilized.  Do not bring them
back and forth.  Organisms from the air can be particularly robust sources of
contamination.

That book will take you step by step and explains how the enthusiest can do
it....if your doing tissue culture i think your an enthusiest. The most
difficult thing I had was keeping things sterile. A small room like a wash
room where you can control the lights, humidity, and cleanliness easier
helps alot. An aquarium will work but you will get elotation from sunlight.
I did cactus and agave and had great results. I would also look at the kits
on http://www.hometissueculture.org/ thats wat i used to get started.
